Structural Biochemistry

教育目標 Course Target

1. Structure and ionization properties of amino acids and polypeptides. Ionization equations and calculation of the net charge of polypeptides. 2. Primary, secondary and tertiary structures of proteins, the Ramachandran angles(dihedral angles) of peptides. Concepts of receptors, receptors ligand interactions, enzyme-substrate interactions. The Scatchard equation. 3. Importance of enzyme inhibitors and applications in modern drug developments. Derivation of the equations for competitive enzyme inhibitors and non-competitive enzyme inhibitors. Important applications of new chemotherapeutic agents 4. Enzyme catalytic principles and mechanisms: Aconitase, Carboxypeptidase, DNase I, Ribonuclease. Coenzyme and mechanisms: NADH (with X-ray structure of the enzyme-coenzyme complex), Biotin, Vitamin B12 5. DNA structure, major groove and minor groove figures. DNA-protein interactions, DNA-small ligand interactions. Restriction endonuclease mechanisms.
